All public transport (bus and share taxi) will go from Sihanoukville to Siem Reap via Phnom Penh. The only way to lop a little off would be to go to Battambang first in which case you could (theoretically) avoid having to go right back into Phnom Penh.

Using Phnom Penh Sorya Bus company (near Sorya market):
PP - Sihanoukville, $4-5USD, 3-4hrs
Sihanoukville - Siem Reap, goes via PP.
Bus 1 Sihanoukville - PP, $4-5USD, 3-4hrs.
Bus 2 PP - Siem Reap, $4USD, 5hrs
We did a similar route, leaving Kampot at approx 7am and getting to Siem Reap at approx 6pm.

Is it worth staying in Kep, Kampot and Sihanoukville? I'm a low key traveller and like peace and quiet, any suggestions?

If you want peace and quiet Kep is defo worth a visit, really not a lot to do there apart from eat seafood and relax, maybe take a day trip to the nearby Rabbit Island, which is even more laid back with a nice beach. Accommodation in Kep ranges from $5 to $180.
