Hi, can anyone tell me whether I can visit most of the highlights in Phnom Penh in one day? Or should I spread it out in 2 days? How should I arrange my itinerary?

The key hotspots in Phnom Penh are (in my personal order of importance):

Tuol Sleng (S21)
National Museum
Royal Palace
Killing Fields
Sunset boat trip
Russian market

While you could cram those into a day, I'd say two days would be a far more comfortable approach -- especially if you want to see the Killing Fields. Something like:

Day one:
Morning: Tuol Sleng (S21)
Afternoon: National Museum
Late afternoon: Sunset boat trip

Day two:
Morning: Royal Palace
Morning: Russian market
Afternoon: Killing Fields

If you've got the time, then an extra day in Phnom Penh is worth it. One of my favorite things to do there was just wander around and discover the city. I particularly liked the areas along waterfront and out near the big monument, where there are chances to interact with locals and discover a bit of Cambodian culture and day-to-day life. Have fun. Yes, they accept USD everywhere. You HAVE to pay in USD for the larger amounts. For the smaller transactions you can pay in riel or in USD, but remember that they will charge a bit more. I mean if you change USD to riel, you will get let's say 4100 riel for 1 USD. If you have to pay 4000 riel in USD you will pay 1 USD (difference = 100 riel per USD).
