I don't believe you can get a bus directly from siem reap to kep, the last stretch from Sihanoukville to Kep isn't particularly well services by the bus companies if memory serves. So you'd have to catch a bus from siem reap through phnom penh to sihanoukville, at sihanoukville you'd have to hire a taxi probably in order to get down the coast to kep.

The bus part shouldn't be a problem there are regular services from siem reap to sihanoukville although most will leave between 7-10 in the morning. The taxi you'll need to negotiate, random off the top of my head figure might be in the $40-50 range, obviously better if you can share. Failing that you'd have to stay the night in sihanoukville and see if there are any mini bus services to Kep.

So yes you can, but it probably wouldn't be cheap and it would be a pretty long day.

that wouldn't be a fun day! we travelled SR to PP by bus (big express bus) and then PP to Kep by bus. The second bus was a long, slow, bumpy trip through not so great roads. It was about 5 hours although they said 3 hours. So ... because the trip from SR-PP is only about 5-6 hours (sorry, memory is a bit fuzzy) if you left really early you could probably get from SR to Kep in one day, but it would be an ugly trip. check whether any buses leave PP for Kep in the afternoon to be sure. Any GH or travel agent in SR can probably tell you or check times for PP or Kep and book in advance if you want. although i'd be inclined to wait until you get to PP, in case the bus is late and you have to stay overnight.
also, keep in mind that depending on what bus you take from SR to PP, you probably won't get dropped off at the bus station that the Kep bus leaves from. You may need a taxi from the SR bus to get to the main bus station.

We did it in December 2009 as follows:
- Standard bus from Siem Reap to Phnom Penh . Sold by the Golden Temple Villa guesthouse for $5, pick up at 7am, departure at 8am. It took 6 hours and it dropped us off somewhere by the river in PP. You can also take the slightly more comfortable Mekong Express bus for $11.
- In Phnom Penh we took a Tuk-Tuk to the central station on the basis that there might be a bus to Kep. From the Central Station (by the central market) there are 2 a day: 7.15 and 13.15 to Kep.
- Having missed these buses, we took a Tuk-Tuk to the market where the private taxis go from: Psar Dang Kor (near the stadium - 15mns from Central Market). There's a bunch of Toyota Camrys parked on the pavement and they'll quote you $50 for the taxi to Kep. We managed to grab a minibus headed there with cargo and paid $35 for the ride (3 of us). It took 3 hours to Kep.
