your options are to take the Govt. Bus, or mini-bus. The mini-bus option is the easiest to organize primarily because you can arrange for it at any tour operator in phuket one day in advance and they speak english and they're motivated because they're earning a commission. it costs about 500 or 600 baht/person.
The mini-bus will take you to Dong-Sak, from there you can take a ferry to koh samui. The tour shop in phuket can organize all that for you.

hi Angela,
it seems there are still flights available but not direct, you'll have a stop in Bangkok.
Prices from $295 with Thai Airways.
Taking a bus is not that bad, as Greg mentioned you can take a big bus or a mini-bus.
A big bus is perhaps more comfortable.

300km bus ride from Phuket town to Donsak should take around 5 hours. Since you're going during the peak tourist period I'd try to hook up a minvan ride via your hotel in Phuket and make sure they take you all the way to the Donsak pier. If you end up on a tourist bus do not store any valuables in your big bag/suitcase which gets stored down below. Many reports of crawling monkeys in the baggage compartment. Keep valuables with you! Have a nice time.
