Me and my wife are planning to visit Angkor Wat in October and then spend a week snorkeling in Thailand - Ko Tao. Can anyone advice on the best route and mode of transportation.

We've got two stories on Travelfish which explain all the logistics (though you'll need to read the Bangkok to Siem Reap one backwards).
Bangkok to Siem Reap
Bangkok to Ko Samui, Ko Pha Ngan and Ko Tao
Hope that helps!

bigdaddy, fastest way is fly from SiemReap to Bangkok (Bangkok Airways) then from Bangkok to Samui (Bangkok Airways)and take boat to Koh Tao. If you plan to return homeland from Bangkok, you can buy Discovery Airpass for 3 flights: SR-BKK, BKK-Samui and Samui-BKK. It will be cheaper. See www.bangkokair.com/discoveryairpass/
Alternatively, you can travel overland:
- bus/taxi from SiemReap to border (Poipet)
- bus to Bangkok
- bus+boat to Koh Tao, or, better overnight train + boat
By the way, do you know that weather conditions on Koh Tao in October not so good?
