Hi, I would like to go from Ayutthaya to Sukhothai, what are my options? Also would 1 day in each city be sufficient to see most things there?

Bus if you want to go direct to Sukhothai, but if you're happy to go via Phitsanulok, then the train to there, then bus is a popular choice.
One day in each is sufficient to cover the highlights, but many opt for two days in Sukhothai -- mainly because it is a pleasant place to hang out and there's a few outlying sites worth considering.

Thanks for the info, also how long does the bus directly to Sukhothai take? I think I've read somewhere that there's an overnight bus? Is that true?

The bus is about 4 1/2 - 5 hours.
Rent a motorbike in Sukhothai. They are cheap and a great way to get around.
