Any suggestions? Flights seem pricy online and require overnight stays on the way. Has anyone hired a car/driver and have a cost estimate? Any other options for 2 people willing to pay a bit more than a bus for a direct trip? Thanks!

If you were to drive it you'd need to overnight in at least Saigon, and I'm not sure if a taxi would cross the border for you -- you may need to change vehicles at the border -- not sure.
Time wise, if you left Siem Reap at the crack of dawn you should be able to get to Saigon by early evening -- assuming a smooth transition at the border (that's a big assumption). Then from Saigon to Nha Trang is a good ten to twelve hours driving.
