We recently travelled this route and wanted to share our experience in case it helps anyone else. We booked with Joyous Journeys and contacted them through their Facebook page (JJ-express highway bus). They replied within the hour and gave us all the info.
Tickets cost $19 per person for a comfortable air con bus - the seats are 2-1 format so pretty wide and with good leg room (we are 5"11 and 6"2 so this is always important!). While the bus has no toilet it stopped twice along the journey at very clean well equipped service stations - western toilets. There were also plenty of places to buy food/drinks and ATMs also. Open all hours.
The bus had an English speaking attendant for the journey who came around and offered us each a snack (mochi bun & cake) along with water and a soft drink. You get a blanket on your seat but be prepared for the bus to be cold! Ours was like a fridge and even though everyone mentioned it to the attendant it stayed warmer only for a bit and we wished we'd worn warmer clothes!
Although we didn't get much sleep we'd still do the journey again. The road is quite bumpy in parts so you might struggle if you're light sleepers like us.
It left Yangon at 8pm and arrived around 5am to the bus station about 7km out of Bagan. The taxi drivers will try and convince you to go and watch sunrise right away and although not pushy were a bit in our faces after a night of no sleep. We paid 10,000 kyat for a taxi straight to our guesthouse in Nyuang U - we read after that it should've cost less and that we should have walked a bit to find an honest taxi driver but hey ho...
